Mice deficient in band 3 have profound anemia and reduced perinatal survival, normal cytoskeletal assembly, and markedly reduced membrane stability.

J Clin Invest 1996, 97:1227-1234. This study provides in vivo evidence for the role of the Gardos channel in promoting dehydration of sickle erythrocytes. It also demonstrates the efficacy of oral administration of clotrimazole in preventing sickle cell dehydration.

Because these compounds are nonactive as inhibitors of cytochrome P450, it also demonstrates that this system is not involved in the regulation of the activity of the human erythrocyte Gardos channel.
