Comparison of four cytokine regimens for mobilization of peripheral blood stem cells: IL-3 alone and combined with GM-CSF or G-CSF

Rosenfeld CS, Bolwell B, LeFever A, Taylor R, List A, Fay J, Collins R, Andrews F, Pallansch P, Schuster MW, et al.: Comparison of four cytokine regimens for mobilization of peripheral blood stem cells: IL-3 alone and combined with GM-CSF or G-CSF. Bone Marrow Transplant 1996, 17:179-183. This study shows that sequential IL-3 and GM-CSF for mobilization leads to delayed neutrophil recovery. Even though the number of patients is small, this observation illustrates how the mobilization strategy can influence the quality and, therefore, the engraftment potential of the mobilized product.

Individual stem cell quality in leukapheresis products is related to the number of mobilized stem cells
-
Breems DA, van Hennik PB, Kusadasi N, Boudewijn A, Cornelissen JJ, Sonneveld P, Ploemacher RE: Individual stem cell quality in leukapheresis products is related to the number of mobilized stem cells. Blood 1996, 87:5370-5378. Important study that underscores the necessity of combining frequency and quality analysis to compare progenitors in mobilized peripheral blood and marrow. Based on CAFC-and colony-forming unit assays, the authors concluded that the total progenitor cell potency in leukapheresis harvests is threefold higher than in bone marrow.

Varas F, Bernad A, Bueren JA: Granulocyte colony-stimulating factor mobilizes into peripheral blood the complete clonal repertoire of hematopoietic precursors residing in the bone marrow of mice. Blood 1996, 88:2495-2501. A study in mice that provides evidence that the clonal repertoire in G-PBMC closely reflects the clonal repertoire in the marrow. Therefore, G-CSF does not appear to preferentially mobilize certain subsets of hematopoietic progenitors in these animals.

Pretreatment of donor mice with granulocyte colony-stimulating factor polarizes donor T lymphocytes toward type-2 Cytokine production and reduces severity of experimental graft versus host disease
-
Pan L, Delmonte J Jr, Jalonen CK, Ferrara JLM: Pretreatment of donor mice with granulocyte colony-stimulating factor polarizes donor T lymphocytes toward type-2 Cytokine production and reduces severity of experimental graft versus host disease. Blood 1995, 86(12):4422-4429. The authors show in a murine transplantation model that the T-helper phenotype of the donor after treatment with G-CSF is polarized toward a type II cytokine response. This demonstrates the potential of functional alterations in accessory cell function after cytokine treatment, which may be relevant for graft-versus-host disease and engraftment.
