The amino terminus of apolipoprotein B is necessary but not sufficient for microsomal triglyceride transfer protein responsiveness
-
100 are secreted in the absence of MTP as HDL density particles. Coexpression of MTP enhanced the number of particles secreted but did not change the density. Removal of the amino terminus from various segments of apoB covering the entire polypeptide blocked secretion regardless of MTP coexpression, indicating that the amino terminus of apoB is necessary but not sufficient for MTP responsiveness.

100 as a lipoprotein particle. The authors concluded that disruption of the disulfide bonding of the amino terminus of apoB by dithiothreitol blocks formation of the lipid-binding domains, rendering them incapable of receiving lipid from MTP causing lipoprotein assembly to halt. Thus the proper folding of the amino terminus of apoB is critical for the proper folding of the subsequent sequences.

50 of 0.3 μmol/l. No effect on lipid synthesis was detectable. Pulse-chase analyses showed that although apoB synthesis was inhibited it did not accumulate in the cells. This was attributable to increased cotranslational or immediate posttranslational degradation of apoB. The effect of the inhibitor on apoB degradation could be blocked with dithiothreitol but not N-acetyl-leucyl-leucyl-norleucinal.

Demonstration of a physical interaction between microsomal triglyceride transfer protein and apolipoprotein B during the assembly of apoB-containing lipoproteins
-
Wu X, Zhou M, Huang LS, Wetterau J, Ginsberg HN: Demonstration of a physical interaction between microsomal triglyceride transfer protein and apolipoprotein B during the assembly of apoB-containing lipoproteins. J Biol Chem 1996, 271:10277-10281. This report was the first to provide evidence for a direct physical interaction between MTP and apoB. Immunoprecipitation of hepatoma G2 extracts with an antibody to MTP also precipitated apoB. Pulse-chase analyses showed that this association was detectable only during the first 10 min of chase. Oleic acid treatment increased the extent of association, and the time, while inhibition of triglyceride synthesis with Triacsin D decreased these parameters. Treatment with N-acetyl-leucyl-leucyl-norleucinal to increase intracellular apoB increased the time but not the extent of interaction.

100 in abetalipoproteinemics. The authors concluded that this was because of a block in the translocation of apoB caused by the absence of MTP. This led to truncation of the nascent polypeptide and release of the amino terminus into the lumen of the ER from where it was secreted, albeit inefficiently.
