Utilizing viral marker proteins, which are differentially sorted from each other in polarized cells, this study shows that, in nonpolarized fibroblast lines, the trafficking of basolateral-destined proteins moving to the plasma membrane is different from that of a protein which follows an apical itinerary. This provides the first evidence that the apical- and basolateral-destined routes are also found in nonpolarized cells.

Vesicle budding is BFA-sensitive, inhibitable by N-ethylmaleimide and dependent on PKC.

Preparations of purified clathrin-coated vesicles from MPR-negative cells contain approximately three times less AP-1 than is seen in wild-type MPR-positive fibroblasts. Re-transfection of either the 275kDa MPR cDNA or the 46kDa MPR cDNA increases the level of AP-1 recovered within the purified coated vesicle preparations.

Later, only procathepsin B, which has a high affinity for MPRs, can be retrieved, moved to lysosomes and converted to the mature form while procathepsin L remains within the mature secretory granule population.

    • •], it is shown that assembly of TGN-derived vesicles occurs in at least two distinct stages, one involving the assembly of a coat and a second comprising the vesicle scission reaction. The first, coat-assembly, stage occurs at 20°C; PKC inhibitors block the second, scission, stage but, interestingly, must be added during the first stage to be effective.

Addition of inhibitors of either tyrosine kinase or tyrosine phosphatase activity inhibits the budding of polypeptide-hormone-containing nascent secretory vesicles from the TGN. Using anti-phosphotyrosine antibodies, the phosphorylation of several membrane-associated proteins is indirectly correlated with the inhibition of vesicular transport.
