J Biol Chem 1997, 72:4864-4868. The authors describe the isolation of a cDNA clone encoding a human sulfotransferase that uses glycolipids as acceptor molecules. The gene was only expressed in a human renal cancer cell line and the predicted size of the mature protein (54 kDa) differed remarkably from that described for the mouse brain enzyme (31 kDa). The existence of other enzymes/isozymes involved in brain-specific sulfatide synthesis is discussed.

    • Bosio A, Stoffel W: Functional breakdown of the lipid bilayer of the myelin membrane in central and peripheral nervous system by disrupted galactocerebroside synthesis. Proc Natl Acad Sci USA 1996, 93:13280-13285. This paper, together with Coetzee et al. [40••], reports the generation of a UDP-galactose-ceramide galactosyltransferase (cgt) null mutant mouse. Analysis of this mouse has permitted studies aimed at understanding galactocerebroside biosynthesis and the role played by galactocerebrosides and sulfatides in myelin sheath and oligodendrocyte differentiation, cgt-deficient mice exhibit a severe dysmyelinating phenotype. Reduced conduction velocity of the sciatic nerve as well as the characteristics of compound action potentials recorded from the spinal cord point to a complete loss of the insulation properties (ion impermeability) of the myelin membrane.
