A comprehensive review of a series of experiments performed by these investigators, designed to examine the intracellular trafficking of fatty acids and triglycerides, and the incorporation of triglycerides into the core of newly synthesized lipoproteins in the liver. The authors discuss how intracellular compartmentation of fatty acids contributes to the regulation of hepatic ketogenesis and triglyceride secretion.

Short-term alterations in carbohydrate energy intake in humans. Striking effects on hepatic glucose production, de novo lipogenesis, lipolysis, and whole-body fuel selection
-
Schwarz JM, Neese RA, Turner S, Dare D, Hellerstein MK: Short-term alterations in carbohydrate energy intake in humans. Striking effects on hepatic glucose production, de novo lipogenesis, lipolysis, and whole-body fuel selection. J Clin Invest 1995, 96:2735-2743. This is one of the few studies to attempt to quantitate the contribution of de-novo lipogenesis as a fraction of total hepatic triglyceride fatty acid production in humans. These investigators have shown, using an innovative stable isotope-labelling technique, that carbohydrate feeding stimulates de-novo lipogenesis by approximately 10-fold and spares lipid oxidation; nevertheless absolute hepatic de-novo lipogenesis still accounts for less than 5 g of fatty acids synthesized each day.

Secretion of apolipoproteins A-I and B by HepG2 cells: Regulation by substrates and metabolic inhibitors
-
Kempen HJ, Imbach AP, Giller T, Neumann WJ, Hennes U, Nakada N: Secretion of apolipoproteins A-I and B by HepG2 cells: regulation by substrates and metabolic inhibitors. J Lipid Res 1995, 36:1796-1806. Glucose added to the medium of HepG2 cells increased apoB secretion by inhibiting its intracellular degradation. The stimulatory effects of glucose and oleate on apoB secretion by HepG2 cells was shown to be additive, suggesting that apoB secretion is greatly enhanced when both the internal (via lipogenesis from glucose) and external supply of fatty acids is increased.

J Clin Invest 1996, 98: 2008-2017. Four days of carbohydrate hyperalimentation/hyperinsulinemia in healthy men increased the VLDL-triglyceride secretion rate 3.4-fold. The 2.7-fold increase in secretion rate of preformed fatty acid contributed quantitatively the most to the total VLDL-triglyceride secretion rate, even though de-novo lipogenesis increased greatly.

Exogenous VLDL stimulates apolipoprotein B secretion from HepG2 cells by both pre- and posttranslational mechanisms
-
Wu X, Sakata N, Dixon J, Ginsberg HN: Exogenous VLDL stimulates apolipoprotein B secretion from HepG2 cells by both pre- and posttranslational mechanisms. J Lipid Res 1994, 35:1200-1210. These experiments incubating VLDL with HepG2 cells showed that it is the increase in intracellular triglyceride and not cholesteryl ester that is the critical factor associated with a stimulation of apoB secretion. In addition to the expected posttranslational effects, VLDL also regulated apoB secretion at the pretranslational level.

Interaction between free fatty acids and insulin in the acute control of very low density lipoprotein production in humans
-
Lewis GF, Uffelman KD, Szeto LW, Weller B, Steiner G: Interaction between free fatty acids and insulin in the acute control of very low density lipoprotein production in humans. J Clin Invest 1995, 95:158-166. Insulin was shown in humans to acutely inhibit VLDL-apoB and VLDL-triglyceride production (by about 50%). partly by the acute suppression of plasma FFAs and partly by a non-FFA mediated (probably direct hepatic) effect. An acute elevation of FFAs stimulated VLDL-triglyceride and VLDL-apoB production.
