Hippocampal mossy fiber sprouting and synapse formation after status epllepticus in rats: Visualization after retrograde transport of biocytin

J Comp Neurol 1995, 352:515-534. Retrograde labeling with biocytin after chemoconvulsant-induced status epilepticus in rats demonstrates recurrent granule cell-granule cell synapses in the dentate molecular layer. These results are the strongest evidence to date for the formation of a recurrent excitatory circuit following seizure-induced hippocampal injury.

Basal expression and induction of glutamate decarboxylase and GABA in excitatory granule cells of the rat and monkey hippocampal dentate gyrus

J Comp Neurol 1996, 373:593-618. This study used immunocytochemical methods to provide evidence that DGCs are capable of synthesizing GABA, and that this activity is enhanced in the perforant path stimulation seizure model. These intriguing findings raise the possibility that DGCs may be capable of inhibitory neurotransmission as a compensatory mechanism in the epileptic state.

The limbic system-associated membrane protein is an lg superfamlly member that mediates selective neuronal growth and axon targeting
-
Pimenta A, Zhukareva V, Barbe M, Reinoso B, Grimley C, Henzel W, • Fischer I, Levitt P: The limbic system-associated membrane protein is an lg superfamlly member that mediates selective neuronal growth and axon targeting. Neuron 1995, 15:287-297. LAMP has previously been shown to be expressed in cortical and subcortical neurons within the 'limbic' system (including DGCs), and is specifically located on growth cones and axons during pathway formation in early development. In this study, the authors demonstrate that intrahippocampal injection of an anti-LAMP antibody during the second postnatal week of the rat results in aberrant growth of mossy fibers.

Lowenstein D, Arsenault L: Dentate granule cell layer collagen expiant • cultures: spontaneous axonal growth and induction by brain-derived neurotrophic factor or basic flbroblast growth factor. Neuroscience 1996, 74:1197-1208. This study demonstrates the feasibility of growing microdissected regions of the dentate granule and pyramidal cell layers within a 3-dimensional collagen matrix. The cultures allow for direct, real-time visualization of growing axons into the matrix, and therefore provide a system for the study of axon guidance molecules on developing mossy fibers.

A nerve growth factor peptide retards seizure development and inhibits neuronal sprouting in a rat model of epilepsy
-
Rashid K, Van der Zee CE, Ross GM, Chapman C, Stanisz J, Riopelle R, • Racine R, Fahnestock M: A nerve growth factor peptide retards seizure development and inhibits neuronal sprouting In a rat model of epilepsy. Proc Natl Acad Sci USA 1995, 92:9495-9499. Consistent with the findings in their previous study [36] the authors show that infusion of a peptide fragment that inhibits binding of NGF to its receptor (TrkA) during kindling delays the development of various stages of seizures. This is associated with a decrease in mossy fiber sprouting, although the effects of the peptide on seizures and sprouting cannot be separated. Importantly, this study demonstrates the potential utility of chronic infusion of function-blocking peptides in studies of epileptogenesis.

Selective inhibition of axon outgrowth by antibodies to NGF in a model of temporal lobe epilepsy
-
Holtzman D, Lowenstein D: Selective inhibition of axon outgrowth by • antibodies to NGF in a model of temporal lobe epilepsy. J Neurosci 1995, 15:7062-7070. This study shows that intraventricular infusion of an anti-NGF antibody attenuates cholinergic but not mossy fiber sprouting following pilocarpine-induced seizures. The approach emphasizes the importance of recognizing that numerous neuronal networks related to the dentate gyrus may be reorganizing during epileptogenesis. The study also demonstrates further the potential value of using in-vivo infusions to address mechanisms of network plasticity.

Overexpression of the neural growth-associated protein GAP-43 induces nerve sprouting in the adult nervous system of transgenic mice
-
Aigner L, Arber S, Kapfhammer J, Laux T, Schneidere C, Botteri F, Brenner • HR, Caroni P: Overexpression of the neural growth-associated protein GAP-43 induces nerve sprouting in the adult nervous system of transgenic mice. Cell 1995, 83:269-278. The authors produced transgenic mice that have persistent expression of GAP-43 at levels similar to the transient peak normally seen during development. Besides causing prominent sprouting of motor neurons at the neuromuscular junction, overexpression of GAP-43 was associated with sprouting of mossy fibers in stratum oriens of CA3. Interestingly, the same transgenic mice with enhanced mossy fiber sprouting had spontaneous seizures, but the sprouting was still seen when animals were treated with phenobarbital. This paper is part of a growing body of literature describing the use of transgenic technology to study neuronal network plasticity.
