In this study, the authors demonstrate that intrahippocampal injection of an anti-LAMP antibody during the second postnatal week of the rat results in aberrant growth of mossy fibers.

Consistent with the findings in their previous study [36] the authors show that infusion of a peptide fragment that inhibits binding of NGF to its receptor (TrkA) during kindling delays the development of various stages of seizures. This is associated with a decrease in mossy fiber sprouting, although the effects of the peptide on seizures and sprouting cannot be separated. Importantly, this study demonstrates the potential utility of chronic infusion of function-blocking peptides in studies of epileptogenesis.

The approach emphasizes the importance of recognizing that numerous neuronal networks related to the dentate gyrus may be reorganizing during epileptogenesis. The study also demonstrates further the potential value of using in-vivo infusions to address mechanisms of network plasticity.

Cell 1995, 83:269-278. The authors produced transgenic mice that have persistent expression of GAP-43 at levels similar to the transient peak normally seen during development. Besides causing prominent sprouting of motor neurons at the neuromuscular junction, overexpression of GAP-43 was associated with sprouting of mossy fibers in stratum oriens of CA3. Interestingly, the same transgenic mice with enhanced mossy fiber sprouting had spontaneous seizures, but the sprouting was still seen when animals were treated with phenobarbital. This paper is part of a growing body of literature describing the use of transgenic technology to study neuronal network plasticity.
