Increased numbers of lymphocytes, eosinophils and mast cells associated with intracellular staining for IL-3, IL-5 and GM-CSF are detected in the gut mucosa of asthmatic and atopic patients without gastrointestinal symptoms, suggesting that the whole mucosal immune system is involved in bronchial asthma.

In order to investigate the hypothesis that eosinophils cajse M2 muscarinic receptor dysfunction, ovalbumin-sensitized guinea pigs were treated with TRFK-5, a monoclonal antibody to IL-5, before an aerosol challenge with ovalbumin. In antibody-treated animals the migration of eosinophils into the lungs was inhibited and the M2 receptor function was normal. In contrast, the function of M2 muscarinic autoraceptors on parasympathstic nerves was decreased in the lungs of untreated challenged animals. The function of M2 muscarinic receptors can thus be protected by inhibiting eosinophil influx into the lung.

Eur J Immunol 1995, 25:245-251. Airway inflammation was induced by intratracheal challenge wild a parasite antigen (egg antigen of Schistosoma mansoni) in presensitized mice. A significant increase of MIP-1α mRNA and protein expression was detected after challenge. MIP-1α was immunolocalized in macrophages and eosinophils present in the bronchoalveolar lavage fluids. Treatment with neutralizing MIP-1α antibodies revealed a specific decrease in eosinophil recruitment to the lungs and airways.

Human eotaxin is directly chemotactic for eosinophils but not for mononuclear cells or neutrophils. It is an early response gene of cytokine-stimulated epithelial and endothelial cells, and is induced in eosinophils by IL-3. Eotaxin mRNA is particularly expressed in the lesions of patients with inflammatory bowel disease.

Immunostaining procedures revealed the existence of IL-4-positive eosinophils in nasal polyp tissues and, to a lesser extent, in bronchial tissues, both from asthmatic patients. Stimulation of purified blood eosinophils with slgA immune complexes induced expression of IL-4 mRNA and the release of IL-4 into the supernatants. Eosinophils can be considered as a primary source of IL-4 in tissues during mucosal inflammation.

IL-4 mRNA was detected by reverse transcription-polymerase chair reaction in blood eosinophils from atopic asthmatics. By enzyme-linked immunosorbent assay, IL-4 was quantified in whole cell extracts and 30% was released in the supernatants from activated cells. Immunocytochemistry revealed that IL-4 is located in the crystalloid of eosinophil granules. Eosinophils can thus contribute to the development of the Th2 phenolype as well as to IgE synthesis.

Reverse transcription polyrnerase chain reaction and Southern blot hybridization allow the detection of FcεRI α-chain mRNA in purified blood eosinophils from four out of five patients with allergic rhinitis; expression was increased by IL-4. Flow cytometry or immunostaining did not reveal the expression of FcεRI on peripheral blood eosinophils; however, some ECP-positive cells expressed FcεRI in the nasal mucosa. These data confirm that FcεRI could be synthesized by eosinophils themselves and that its expression is correlated with eosinophil activation.

Eur J Immunol 1995, 26:117-125. Flow cytometry and various binding procedures showed the existence of a specific receptor lor SC on purified human eosinophils. This 15kDa molecule binds to SC or slgA but not to serum IgA, and can mediate eosinophil degranulation induced by SC or slgA immune complexes. The existence of such a receptor might explain the preferential degranulation induced by slgA, and strengthen the probable involvement of eosinophils in mucosal immunity.

In order to examine the role of the physiological stimuli that trigger ensinophils to generate cytokines, the authors show that C5a and FMLP but not platelet-activating factor or C-C chemokines (RANTES, MIP-1α, MIP-1β) induce the release of IL-8 and GM-CSF by eosinophils, but only in the presence of cytochalasin B.
