    • Song Z, Casolaro V, Monos D, Ono SJ: Identification of polymorphic nucleotides within the human interleukin-4 gene promoter which mediate overexpression of the gene. J Immunol 1996, 156:424-420. Demonstration that polymorphic nucleotides with the human IL-4 proximal promoter can affect promoter activity by altering the affinity of DMA-binding proteins for both positive regulatory element and negative regulatory element sequences. The electrophorectic mobility-shift assay alteration stabilizes AP-1 interaction near the P-sequence, an essential event in TCP-mediated activation of cytokine gene expression.

    • Abnormal IL-4 gene expression by atopic dermatitis T lymphocytes is reflected in altered nuclear protein interactions with IL-4 transcriptional regulatory elements
    • Ghan SC, Brown MA, Willcox TM, Li SH, Stevens SR, Tara D, Hanifin JM: Abnormal IL-4 gene expression by atopic dermatitis T lymphocytes is reflected in altered nuclear protein interactions with IL-4 transcriptional regulatory elements. J Invest Dermatol 1996, 106:1131-1136. Clear evidence from in situ hybridization analysis that atopic dermatitis patients have a higher frequency of IL-4-producing peripheral blood T cells when compared to normal subjects. By electrophorectic mobility-shift assay, the investigators show that nuclear extracts from twelve out of twelve patients with atopic dermatitis form an activation complex on the IL-4 promoter, compared to two out of twelve normal subjects. This seminal discovery suggests that the genes encoding IL-4 promoter binding proteins may themselves by atopy genes.

    • Lederer JA, Liou JS, Kim S, Rice N, Lichtman AH: Regulation of NF-kappa B activation in T helper 1 and T helper 2 cells. J Immunol 1996, 156:56-63. An extremely important publication indicating that TCR stimulation fails to induce p65 (RelA) in Th2 cells. The paper also shows that Th1 and Th2 cells express different levels and/or different forms of IκBα, and that cytokines (but not TCR stimulation) modulate cytoplasmic IκB. The implications of the paper are that cytokines can modulate the Rel response to TCR stimulation suggesting that Rel proteins might contribute to Th differentiation. Indeed that Rel knockout mice do have abnormalities in Th development and cytokine production.
