Clear evidence from in situ hybridization analysis that atopic dermatitis patients have a higher frequency of IL-4-producing peripheral blood T cells when compared to normal subjects. By electrophorectic mobility-shift assay, the investigators show that nuclear extracts from twelve out of twelve patients with atopic dermatitis form an activation complex on the IL-4 promoter, compared to two out of twelve normal subjects. This seminal discovery suggests that the genes encoding IL-4 promoter binding proteins may themselves by atopy genes.

An extremely important publication indicating that TCR stimulation fails to induce p65 (RelA) in Th2 cells. The paper also shows that Th1 and Th2 cells express different levels and/or different forms of IκBα, and that cytokines (but not TCR stimulation) modulate cytoplasmic IκB. The implications of the paper are that cytokines can modulate the Rel response to TCR stimulation suggesting that Rel proteins might contribute to Th differentiation. Indeed that Rel knockout mice do have abnormalities in Th development and cytokine production.
