of outstanding interest. An important study utilizing soluble, purified preparations of the main components of the TCR-MHC/peptide interaction to explore the kinetics and affinity of binding of a well characterized TCR to MHC/peptide complexes. The authors provide the strongest evidence which directly correlates the affinity of the interaction between the TCRs and MHC/peptide ligands with the outcome of thymic selection

Mechanisms of acquired thymic tolerance in experimental autoimmune encephalomyelitis: Thymic dendritic-enriched cells induce specific peripheral T cell unresponsiveness in vivo
-
in vitro with an encephalitogenic peptide and injected intravenously were capalbe of preventing the development of EAE but this protection was lost in thymectomized recipients. Furthermore, the intravenous injection of thymic dendritic cells, isolated from animals previously injected intrathymically with the peptide, also prevented the development of EAE. These findings suggest that an interaction of thymic dendritic cells with a specific TCR of activated peripheral T cells can lead to the inactivation of the antigen-specific cells
