Quail anterior endoderm was co-cultured with HH stage 4 chick posterior primitive streak cells that do not form cardiac muscle normally. Commitment of chick cells to the myocyte lineage was assayed using three independent muscle markers. A decrease in globin mRNA in the same cells suggested that the quail endoderm induced a change in fate from the hematopoietic to the cardiac muscle lineage.

Explant cultures of Xenopus embryos show that deep dorsoanterior endoderm can induce cardiogenesis in the absence of Spemann's organizer at stage 10 (early gastrula). Both the organizer and the dorsoanterior endoderm are required to induce cardiogenesis in non-muscle forming regions of the embryo. These data suggest that the mechanism of heart formation is conserved among vertebrates.

Development 1995, 121:3889-3899. XNkx-2.3, a Xenopus homologue of mouse Nkx-2.5 [34], is expressed in the heart in a pattern that suggests it plays an important role in normal cardiac myogenesis. Perhaps Nkx-2.3 is a functionally redundant nuclear factor that prevented the Nkx-2.5 null mouse [36••] from exhibiting a more severe phenotype (i.e. the lack of a simple heart tube).

This report (and [47,48]) show that two GATA-4 homologues, GATA-5 and GATA-6, are likely to play important roles in normal cardiac myogenesis. These data provide further support to the hypothesis that the mechanism of heart formation is conserved among vertebrates.

Development 1996, in press. This paper and [63] document a large number of mutations in the zebrafish that affect cardiac morphogenesis and function. Although the mutated genes have not yet been identified, this approach promises to identify a number of novel genes that play central roles in vertebrate heart development.
