Blood 1995, 86:2534-2540. This French group used oligonucleotide primers that correspond to the conserved WSXWS motif found in the hematopoietic cytokine receptor family to identify cDNA that encodes the human interleukin-11 receptor. The human interleukin-11 receptor nucleotide sequence is highly homologous (82%) to that of the previously cloned murine interleukin-11 receptor. An interleukin-11 receptor isoform that lacks a cytoplasmic domain was identified, supporting the view that gp130 is the signal-transducing subunit in the receptor complex.

In the phase I study of recombinant human interleukin-11 in patients with breast cancer, all subjects had increases in platelet counts after treatment with recombinant human interleukin-11. In addition, after moderately high-dose chemotherapy (cyclophosphamide and doxorubicin), patients receiving recombinant human interleukin-11 at doses of 25 to 75 μg/kg had higher nadirs than patients treated with 10 μg/kg. Treatment was well tolerated, with anemia being the most common side effect. Other studies have shown that this anemia is due to an increase in plasma volume, not a decrease in erythrocyte mass. The first randomized, placebo-controlled study that has shown a decrease in the number of patients with severe chemotherapy-induced thrombocytopenia who required platelet transfusions during their next chemotherapy cycle. The patients in this study had already been transfused platelets during a previous cycle because of severe chemotherapy-induced thrombocytopenia and they continued to receive the same chemotherapy (with no dose reductions) during the study. In patients receiving recombinant human interleukin-11, there also were trends toward fewer platelet transfusions and more rapid recovery to platelet counts of 50,000 and 100,000 cells/μL compared with patients in the placebo group.
