Virology 1996, 216:317-325. This is the first direct demonstration that the NS5 protein alone can serve as the RNA-dependent RNA polymerase for dengue viruses. This advance could make it easier to investigate factors involved in the regulation of dengue virus replication and to screen for drugs that inhibit polymerase activity.

Two amino acid substitutions were made in the dengue 3 part of the chimeric dengue 3-dengue 4 virus constructed in this study. One of these changes, the substitution of lysine for glutamine at amino acid site 406 in the envelope gene, led to the acquisition of mouse neurovirulence in the mutant chimera.

    • Characterization of a highly attenuated Japanese encephalitis virus generated from molecularly cloned cDNA
    • Sumiyoshi H, Tignor GH, Shope RE: Characterization of a highly attenuated Japanese encephalitis virus generated from molecularly cloned cDNA. J Infect Dis 1995, 171:1144-1151. Six full-length cDNA clones of the Japanese virus genome were constructed by in-vitro ligation of different cDNA fragments containing the 3′ or 5′ halves of the genome. Three of the recombinant viruses produced from the full-length cDNA clones by RNA transcription were attenuated for mice. One of the attenuated strains was shown to have a single point mutation, the substitution of lysine for glutamic acid at amino acid site 138 in the envelope protein. This recombinant virus remained highly immunogenic, but no longer killed mice either by the intracranial or subcutaneous routes.

    • + cytotoxic T lymphocyte (CTL) clones. Recognition of NS3 amino acids 500 to 508 by CTL clones of two different serotype specificities
    • + cytotoxic T lymphocyte clones established from a dengue 4-immune adult exhibited three patterns of reactivity: dengue 4-specific, crossreactive for dengue 2 and dengue 4, and crossreactive for all four serotypes of dengue virus. Several of the dengue 4-specific and one of the dengue 2-dengue 4 crossreactive clones were shown to recognize the same 9 amino acid peptide on the dengue 4 non-structural protein NS3, but the crossreactive clone used a different T cell receptor. These cytotoxic T lymphocyte clones were shown to be restricted by the HLA-B35 allele.

    • + cytotoxic T lymphocyte clones were established by stimulating peripheral blood mononuclear cells from a dengue 4 immune donor with inactivated dengue 4 viral antigens. Seven of these clones were shown to recognize the dengue 4 capsid protein. A dengue 4-specific clone was shown to recognize an epitope between amino acids 47-55 of the capsid protein and several clones that reacted with both dengue 4 and dengue 2 recognized a separate epitope between amino acids 83-92.

In 1993 there were 57 cases and in 1994, 91 cases of dengue reported to state and federal health agencies. Cases occurred in American tourists who had travelled to the Caribbean islands, Mexico, Central America, Asia and Africa. Six patients were hospitalized, three with syndromes resembling DHF.

Am J Trop Med Hyg 1995, 53:89-94. Fifty-nine of 129 febrile troops acquired dengue in Somalia less than 1 month after arrival. Although dengue is transmitted by a daytime-biting mosquito, the only positive risk factor for dengue infection was the failure to use bed nets. Most troops failed to take anti-mosquito precautions such as the use of insect repellents or the wearing of long-sleeved shirts.

Abnormal thickening of the gallbladder wall was observed in 93.8% of children with DSS. This observation introduces a simple, rapid, non-invasive outpatient test which identifies serious vascular permeability and a need for immediate hospitalization and fluid therapy.

Sixty-eight per cent of cases and 47% of controls received no vaccine. Effectiveness for one dose was 61% and for two doses 97.5%.
