A comprehensive study that showed that oral immunization of mice with as little as 5 μg of recombinant H. pylori urease, together with E. coli LT as the mucosal adjuvant, was sufficient to prevent H. felis infection in the animals. Moreover, the authors investigated systemic and local antibody responses with respect to protection status and reported that secretory IgA levels correlated with protection.

This letter reports on a double-blind phase I clinical study in which 12 healthy adults with asymptomatic H. pylori infection were given either 60 mg of recombinant H. pylori urease or placebo once weekly for 4 weeks. Gastroscopy was performed at the beginning of the trial and 1 month after the final dose of urease or placebo. No significant differences in gastric disease were observed between the two groups, suggesting that oral immunization with urease was well tolerated. These results have fundamental importance for the development of an H. pylori vaccine.

Gastroenterology 1995, 109:115-121. These authors present data that extend the findings of previous studies of therapeutic immunization using whole-cell sonicates. In this paper, it was shown that immunization with the UreB subunit of H. pylori urease and cholera toxin was sufficient to clear H. felis infection in 56% of mice and that clearance was accompanied by the absence of body gastritis. Significantly, 9 of 14 mice in which the infection had been eradicated were immune to a subsequent challenge.

Proc Natl Acad Sci U S A 1995, 92:6499-6503. In this study, the HSP homologues of H. pylori (HspA and HspB) were shown to be protective antigens in the H. felis mouse model. This was the first report in which a protein belonging to the GroES family (H. pylori HspA) was shown to be effective as a protective antigen. It was also shown that coadministration of HspA with H. pylori UreB provided a protective efficacy statistically comparable to that of whole-cell sonicates. Induction of protective immunity appeared to correlate with consistently high antigen-specific IgG1 responses in the sera of mice.

The authors reported that the severity and type of pathologic disorders observed in the infected mice varied according to the genetic background of the host. The data supported the suggestion that host properties are major contributing factors in the outcome of Helicobacter infection.
