This publication demonstrates that tissue factor-induced coagulation is required for metastatic dissemination of melanoma cells in a hematogenous metastasis assay. Inhibition of coagulation at the level of tissue factor, factor Xa, and thrombin abolished metastasis in vivo. Thrombin, but not factor Xa, provided a proliferative signal to the tumor cell, a possible mechanism by which tissue factor expression supports tumor cell survival in vivo.

This translocation of tissue factor results in downregulation of tissue factor function but also positions the receptor into a cellular compartment that is enriched in signaling molecules.

Endothelial cells in proximity to the tumor also express tissue factor, providing evidence in human tumors that induction of tissue factor and the angiogenic response are spatially correlated.

The deletion of the cytoplasmic domain of tissue factor resulted in the loss of the prometastatic properties of tissue factor. This article thus provides strong evidence that tissue factor supports metastasis through cellular functions that go beyond the activation of the coagulation cascade.
