To show the direct effect of apolipoprotein E on atherogenesis in the vascular wall, transgenic mice expressing human apolipoprotein E under control of the H2 Ld promoter were established. Transgenic mice with high expression of the human apolipoprotein E in the arterial wall demonstrated reduction in the formation of fatty streak lesions through enhanced cholesterol efflux from the aortic wall into plasma.

To study the role of apolipoprotein E on plasma cholesterol metabolism, bone marrow transplantation was performed on apolipoprotein E deficient mice with wild-type mouse bone marrow cells. The results demonstrated an important contribution of macrophage-derived apolipoprotein E both in reducing plasma cholesterol levels and in preventing the progression of atherosclerosis.

Deletion analysis of the c-fms promoter revealed that a region including a binding site for transcription factor PU.1 was required for transcriptional activity in human aortic medial smooth muscle cells.
