Hurt A nascent pre-ribosome with a tadpole-like structure functions in ATP-dependent maturation of 60S subunits Mol Cell 15 2004 295 301 This is the first structural analysis of a yeast pre-ribosome. Electron microscope images of a late pre-60S complex reveal a striking elongated structure with a tadpole-like morphology due to the presence of the huge (560 kDa) Rea1p AAA-type ATPase. ATP treatment of this complex induces dissociation of Rea1p, suggesting that this factor and perhaps the associated Rix1p complex mediates the ATP-dependant remodelling of the pre-60S complex. This is potentially essential for the acquisition of competence for nuclear export.

The levels of polyadenylated rRNAs are increased in strains lacking the nuclear exosome component Rrp6p, and this might reflect a quality-control mechanism of rRNA biogenesis.

Milner Disruption of the nucleolus mediates stabilization of p53 in response to DNA damage and other stresses EMBO J 22 2003 6068 6077 The authors demonstrate that inducing nucleolar disruption causes p53 stabilisation and propose that the nucleolus is a stress sensor responsible for maintenance of low levels of p53. These observations potentially provide a unifying mechanism for the apparently diverse pathways of p53 activation.

    • Ribosomal protein L23 activates p53 by inhibiting MDM2 function in response to ribosomal perturbation but not to translation inhibition
    • •], this paper shows that ribosomal protein (rp)L23, as well as L11, can associate with the ubiqitin ligase Hdm2p (Mdm2p in mice). The data suggest that the levels of rpL23 respond to changing growth conditions, causing p53-dependant cell cycle arrest by inhibiting the HDM2-induced p53 polyubiquitination and therefore degradation. These observations point to the key role of the nucleolus in the cellular stress response.

    • M. Oeffinger, and D. Tollervey Yeast Nop15p is an RNA-binding protein required for pre-rRNA processing and cytokinesis EMBO J 22 2003 6573 6583 The nucleolar protein Nop15p is characterised as a factor required for ribosome biogenesis as well as cytokinesis. Nop15p-depleted cells indeed arrest as large and elongated cells with divided nuclei but undivided nucleolus and fail to assemble a contractile actin ring at the bud neck.
