    • 3H)-CP55,940 (1.5 nM final), 2.5 μl test compound diluted in DMSO (0-10 μM final (10, 3, 1, 0.3, 0.1, 0.03, 0.01, 0.003 μM), and 7.5 μg of membrane protein. Nonspecific binding was determined in the presence of 1 mM cold CP55.940. Reactions were incubated for one hour at 30 degrees, filtered onto GF/B plates (pretreated with 0.3% polyethylenimine) using a Packard Micromate 196, and washed ten times with cold wash buffer (50 mM Tris and 0.5% fatty acid-free BSA). Plates were dried for one hour at 50°C and counted on a topcounter after adding Microscint 0 to each well.
