    • 2. The cells were cultured in high glucose RPMI-1640 media containing 10% FBS, 100 IU/mL penicillin, and 100 μg/mL streptomycin. Cell cytotoxicity was determined by MTT assay. Briefly, cells were seeded in 96-well-plate (5000 cells/well) and incubated for 48 h. Then, the tested compounds with various concentrations were added to the wells and 48 h later the MTT solution (0.5 mg/mL) was added and incubated for 1 h. DMSO (100 μL) was added to each well to dissolve the reduced MTT crystals. The absorbance of each well was measured at 570 nm with a microplate reader.
