Although a true isodichroic point is expected only for a pure two-state transition, strongly cooperative helix-coil transitions offen give apparent isodichroic points. The pH dependency of the CD spectra of CA(1-7)M(2-9) adds to the complexity of the HFIP-induced transition, as HFIP is very acidic due to the effect of the six fluorine groups, and the pH of the solution varies by ca. one pH unit during the titration.

The lack of concentration dependence of the ultracentrifugation data contrasts with the CD results, which show a considerable variation in the same concentration range. This may be due to the different sensitivities of the two techniques or to small differences in pH between samples, which could cause significant differences in the degree of aggregation, since the pH of 6.5 is very close to that at which the onset of precipitation in 5 mM phosphate and 12% HFIP occurs.
