One of a series of recent papers showing important functional associations between integrins and other cell surface receptors. In this paper, the association of CD87 with integrin CR3 (CD11b/CD18) is shown to require prior activation of the integrin, and appears to be indirectly mediated through caveolin, a two-membrane spanner. Formation of a CD87-integrin complex leads to inhibition of integrin-mediated adhesion but facilitates CD87 adhesion to vitronectin.

This study elucidates the role of a conserved triple threonine (TTT) motif in the cytoplasmic tail of CD18 on CD11a/CD18-mediated adhesion. A TTT to AAA substitution had no effect on receptor affinity yet reduced integrin-mediated spreading because of a decrease in the ability of the integrin to form focal adhesions and to organize the cytoskeleton into stress fibers. The authors suggest that the integrin β subunit cytoplasmic tails may be more important in modulating post-receptor binding events than in receptor affinity modulation.

This paper reports the three-dimensional structure of a fragment of fibronectin containing type III repeats 7 to 10, the latter containing the RGD motif. The structure reveals the important interdomain relationships between the RGD loop in domain 10 and the "synergyregion of domain 9 (which has also been implicated in integrin binding). The structure reveals that both regions lie on the same face of the modular FN7-10 structure and are thus easily accessible for binding to a single integrin.
