Regulation of integrin function by the urokinase receptor
-
Wei Y, Lukashev M, Simon DI, Bodary SC, Rosenberg S, Doyle MV, Chapman HA: Regulation of integrin function by the urokinase receptor. Science 1996, 273:1551-1555. One of a series of recent papers showing important functional associations between integrins and other cell surface receptors. In this paper, the association of CD87 with integrin CR3 (CD11b/CD18) is shown to require prior activation of the integrin, and appears to be indirectly mediated through caveolin, a two-membrane spanner. Formation of a CD87-integrin complex leads to inhibition of integrin-mediated adhesion but facilitates CD87 adhesion to vitronectin.

Modulation of cell activation by changes in aLb2 (LFA-1, CD11a/CD18) cytoplasmic domain/cytoskeleton interaction
-
Peter K, O'Toole TE: Modulation of cell activation by changes in aLb2 (LFA-1, CD11a/CD18) cytoplasmic domain/cytoskeleton interaction. Exp Med 1995, 181:315-326. This study elucidates the role of a conserved triple threonine (TTT) motif in the cytoplasmic tail of CD18 on CD11a/CD18-mediated adhesion. A TTT to AAA substitution had no effect on receptor affinity yet reduced integrin-mediated spreading because of a decrease in the ability of the integrin to form focal adhesions and to organize the cytoskeleton into stress fibers. The authors suggest that the integrin β subunit cytoplasmic tails may be more important in modulating post-receptor binding events than in receptor affinity modulation.

A crystal structure of a four-domain segment of human fibronectin encompassing the RGD loop and synergy region
-
Leahy DJ, Aukhil I, Erickson HP: A crystal structure of a four-domain segment of human fibronectin encompassing the RGD loop and synergy region. Cell 1996, 84:155-164. This paper reports the three-dimensional structure of a fragment of fibronectin containing type III repeats 7 to 10, the latter containing the RGD motif. The structure reveals the important interdomain relationships between the RGD loop in domain 10 and the "synergyregion of domain 9 (which has also been implicated in integrin binding). The structure reveals that both regions lie on the same face of the modular FN7-10 structure and are thus easily accessible for binding to a single integrin.

-
Qu A, Leahy DJ: The role of the divalent cation in the structure of the I domain from the CD11a/CD18 integrin. Structure 1996, 4:931-942. Papers by Lee et al. (Cell 1995, 80:631-638 and Structure 1995, 3:1333-1340) and Qu and Leahy (Proc Natl Acad Sci USA 1995, 92:10277-10281 and Structure 1996, 4:931-942) reveal the structure of metal-bound and free forms of the A-domain. These data suggest that the structure exists in different conformations that also affect the topology and electrostatic potential of the region surrounding MIDAS, and therefore the ability of the respective integrin to bind ligands.
