H. Hou, Y. Song, H. Xu, Y. Wei, Y. Fan, Y. Zhu, L. Li, C. Du, Macromolecules 36 (2003) 999. Bis(4-pyridylformyl)piperazine was prepared by a modification of this published procedure. Isonicotinoyl chloride hydrochloride (32.22 g, 0.1810 mol) was added to 250 mL of dry pyridine in a 500 mL round bottomed flask. In a separate 250 mL round bottomed flask anhydrous piperazine (7.755 g, 0.0922 mol) was added to 75 mL of dry pyridine. Both solutions were heated gently until all solid material had dissolved. The piperazine solution was then added to the isonicotinoyl chloride hydrochloride solution. The reaction mixture was refluxed for 10 hours under flowing nitrogen. The original brown color had changed to brown-red, with formation of an off-white polycrystalline precipitate of 4-bpfp. After the reaction mixture was allowed to cool to 25 °C, the 4-bpfp solid was isolated by filtration and washed with cold water followed by cold ethanol (22.084 g, 0.0900 mol, 82.8 % yield). Note: Failure to permit the reactants to dissolve completely in the initial solutions before mixing results in substantially decreased product yield and contamination with piperazine dihydrochloride. If the pyridine solvent is not dried, hydrolysis of the isonicotinoyl chloride hydrochloride precursor results in isolation of isonicotinic acid.
