    • Functional expression cloning of Nanog, a pluripotency sustaining factor in embryonic stem cells
    • ••] identify a novel homeoprotein, Nanog, which is crucial for the self-renewal of ES cells in culture and for the maintenance of pluripotency in inner cell mass cells in vivo. Over-expression of Nanog sustains Oct-4 expression and promotes ES cell self-renewal in the absence of LIF. Nanog works independently of Stat3 but requires Oct-4 expression to maintain pluripotency. The restriction of Nanog expression to pluripotent cells demonstrates that these cells employ some self-renewal mechanisms that are distinct from those used by multipotent stem cells.

BMP signaling inhibits neuronal differentiation by ES cells in culture by inducing Id gene expression, which impairs neurogenesis by inhibiting the function of neurogenic basic helix-loop-helix transcription factors. BMP signaling is necessary to promote ES cell self-renewal in culture because FGF secreted by ES cells causes them to undergo neuronal differentiation (see [17]). This demonstrates that factors that block lineage-specific differentiation can be critical for self-renewal, suggesting that different combinations of such factors may be required by different stem cells, or stem cells in different environments.

Over-expression of Axin, a negative regulator of Wnt signaling, impairs hematopoietic stem cell self-renewal. These data suggest that Wnt pathway activation is capable of promoting hematopoietic stem cell self-renewal. This conclusion is supported by [46], which shows that soluble Wnt protein can promote hematopoietic stem cell self-renewal in culture.
