X-ray crystallographic analysis of ligands from each series cocrystallized with the ligand binding domain of hPPARγ indicates that the carboxylic acid moiety of the N-(2-benzoylphenyl)tyrosines and the 2,4-thiazolidinedione moiety of the TZDs interact with the same portion of the ligand binding domain in the hPPARγ receptor, with the benzophenone occupying a pocket which remains empty in the PPARγ-TZD complex (M. Milburn, unpublished results from these laboratories).
