    • Sox9 directly promotes Bapx1 gene expression to repress Runx2 in chondrocytes
    • In this study Bapx1/Nkx3.2 was identified as a direct target and by using shRNA mediated knock-down of Bapx1 in cultured chondrocytes the authors showed an increase in Runx2 expression. This nicely provides a mechanistic link for previous studies [2,4] and the study by Provot et al. [58]

    • Runx2 inhibits chondrocyte proliferation and hypertrophy through its expression in the perichondrium
    • This in vivo study in the mouse has provided evidence that the interactions of Twist1 and Runx2 are not only involved in regulating osteoblastogenesis, but also important for chondrocyte hypertrophy. The results suggest that Runx2 in addition to promoting hypertrophy through its expression in chondrocytes, negatively regulates hypertrophy by acting in the perichondrium thereby providing an additional level of control in order to coordinate chondrocyte maturation and osteoblastogenesis.

    • NFAT and Osterix cooperatively regulate bone formation
    • This study showed in vivo that inhibition of NFATs using the drug FK506, which inhibits the phosphatase calcineurin an activator of NFAT, leads to reduced bone formation. These data were corroborated by in vitro studies showing that NFATc1 and NFATc2 deficient cells cannot form bone. In vitro binding studies showed that NFATc1 and Osterix interact on the protein level and that this interaction stimulates the transcriptional activity of Osterix thereby providing a potential mechanism of action.

    • Runx2 determines bone maturity and turnover rate in postnatal bone development and is involved in bone loss in estrogen deficiency
    • In this study the authors used a transgenic mouse model expressing dominant-negative form of the transcription factor Runx2 in differentiated osteoblasts and showed that Runx2 activity is not essential to maintain the expression of major bone matrix protein genes in mature osteoblasts, hence raising the question about the in vivo relevance of previous findings that Runx2 is the major regulator of the expression of these bone matrix protein genes in vitro.
