Drosophila exocyst components Sec5, Sec6, and Sec15 regulate DE-cadherin trafficking from recycling endosomes to the plasma membrane
-
This study demonstrates that E-cadherin is recycled from the lateral membrane to the apical membrane in the Drosophila pupal notum. This process requires the exocyst complex subunits Sec5, Sec6, and Sec15. The E-cadherin binding partner β-catenin associates directly with the Sec10 exocyst subunit, suggesting a positive feedback loop in which E-cadherin trafficking is targeted to sites of β-catenin localization.

The Drosophila homolog of the Exo84 exocyst subunit promotes apical epithelial identity
-
This paper shows that the exocyst protein Exo84 is required to deliver Crumbs and E-cadherin from recycling endosomes to the cell surface in the Drosophila embryo. Crumbs is the first protein observed to be mislocalized in Exo84 mutants, suggesting that Crumbs is an apical effector of the vesicle transport machinery required for cell polarity.

Cdc42 and Par proteins stabilize dynamic adherens junctions in the Drosophila neuroectoderm through regulation of apical endocytosis
-
This manuscript demonstrates that Cdc42, Par-6, and aPKC regulate endocytosis in the Drosophila embryo. Apical and junctional proteins become mislocalized in these mutants, and E-cadherin mislocalization results in part from an accumulation of Crumbs in the late endosomes. The authors provide evidence that Cdc42 inhibits endocytosis and is required for multiple steps in vesicle transport.

Modulation of intracellular trafficking regulates cell intercalation in the Drosophila trachea
-
This study provides the first example of patterned E-cadherin trafficking in a morphogenetic process. The Spalt transcription factor and its target dRip11 are necessary and sufficient for high levels of apical Rab11 in the tracheal dorsal trunk. Increased junctional turnover in trunk cells is associated with higher surface levels of E-cadherin and an absence of cell rearrangement.

Genome-wide analysis identifies a general requirement for polarity proteins in endocytic traffic
-
This study describes a genome-wide screen making use of a C. elegans strain expressing a fluorescently labeled yolk protein to report defects in yolk secretion (exocytosis) or uptake (endocytosis). The authors identify 168 genes specifically required for yolk trafficking, including CDC-42, PAR-3, PAR-6, and PKC-3. Further experiments in worms and mammalian cells suggest dual and conserved roles for the Par proteins in endocytosis and recycling endosome function.

This study sheds light on the role of membrane trafficking in cell polarity and adherens junction stability. This study provides evidence that cadherin trafficking is essential for adherens junction stability in neuroepithelial cells of the mouse neocortex. The endocytic protein Numb colocalizes with E-cadherin in recycling endosomes near adherens junctions. shRNA-mediated knockdown of Numb disrupts adherens junctions, suggesting that Numb-mediated trafficking of cadherins is required for adherens junction stability in neuroepithelial cells.

Hexagonal packing of Drosophila wing epithelial cells by the planar cell polarity pathway
-
This study shows that epithelial cells in the Drosophila pupal wing reorganize their neighbor relationships to convert from a disordered to hexagonal array. This process occurs through local junctional remodeling and requires dynamin and components of the planar cell polarity pathway.

Dual function of Src in the maintenance of adherens junctions during tracheal epithelial morphogenesis
-
This paper provides evidence that junctional turnover involves a combination of transcriptional and post-translational regulation in the Drosophila trachea. Src42A and Src64B downregulate E-cadherin surface localization and simultaneously promote E-cadherin trafficking through activation of E-cadherin transcription by β-catenin and its nuclear binding partner, TCF.

Multicellular rosette formation links planar cell polarity to tissue morphogenesis

This paper shows that axis elongation in the Drosophila embryo occurs through a combination of local and higher order cell behaviors. These cell rearrangements are associated with a planar polarized distribution of adherens junction proteins that may influence polarized cell behavior during intercalation.

Localized zones of Rho and Rac activities drive initiation and expansion of epithelial cell-cell adhesion
-
This study shows that E-cadherin microdomains are actively translocated toward the periphery of nascent contacts in cultured mammalian epithelial cells. The authors provide evidence that myosin II is locally activated at the edges of the interface, which powers an outward contraction of the cortical actin bundle. Actomyosin contractility underlies the mobility of E-cadherin microdomains and leads to the expansion of the contact area.

A two-tiered mechanism for stabilization and immobilization of E-cadherin

Using the fluorescence imaging techniques of FRAP and photoconversion, this paper shows that dynamic properties of E-cadherin vary in different regions of the cell surface in the Drosophila embryo. These properties are spatially localized in a process that requires α-catenin and the actin cytoskeleton.

Endocytosis optimizes the dynamic localization of membrane proteins that regulate cortical polarity
-
Using FRAP techniques this study measures rates of diffusion, endocytosis, and targeted exocytosis for active Cdc42 in budding yeast. These physical parameters are combined with a mathematical model to demonstrate that a balance of cortical diffusion, endocytosis, and targeted exocytosis maintains proteins in a dynamically polarized state.
