The authors hypothesized that extensively multipassaged swine MSCs would retain therapeutic potency in an acute myocardial infarction model. After 96 population doubling, there was no evidence of cytogenetic abnormalities. Using immunodeficient (nonobese diabetic/severe combined immunodeficient) mice, they show that a fraction of transplanted MSCs remained at least 4 weeks after transplantation, and left ventricular contractile function was significantly improved in transplanted hearts. Importantly, the study showed that few engrafted MSCs developed cardiomyocyte or endothelial cell markers, suggesting that the benefits of transplantation did not derive from structural contributions of engrafted MSCs.
