The authors used GFP-marked ES cells in co-cultures with chick cardiogenic mesodem, the latter being the normal inducer of the liver, and obtained GFP+ cells that expressed endodermal transcription factors and several early liver-specific genes. This study illustrates the feasibility of using embryonic inductive tissue, rather than synthetic culture conditions, to guide stem cell differentiation.

They also showed that ectopic expression of HNF4α in fibroblast cells is sufficient to induce epithelialization, but insufficient to induce liver-specific genes. Thus, HNF4α must be integrated into the network of other hepatic nuclear factors to promote hepatic differentiation.
