Due to stacking abilities of its two aromatic moieties, adenine and nicotinamide, NAD is folded in water solution (with the distance between the bases of 4-5 Å). Several bacterial reductases bind NAD in such compact (folded) conformation (see PDB entries 1RZ1, 1ZPT, 2BKJ). Other enzymes, such as bacterial NAD kinases, including therapeutically important Mycobacterium tuberculosis enzyme, unfold NAD partially; NAD is bent with the distance around 12 Å between the bases. In majority of NAD-utilizing enzymes, including IMPDH, the distance is around 16-17 Å. This means that NAD must be unfolded into extended conformation to be able to bind.
