This study reported the genome sequence of USA300. The study revealed that USA300 is genetically similar to MRSA strain COL and revealed the presence of arginine catabolic mobile element (ACME) in USA300. The study also emphasized how horizontal transfer from other species of staphylococci may be responsible for significant changes that enhance the increased fitness and pathogenicity of this CA-MRSA strain.

J Infect Dis 2006; 194:1761-1770. This study used human neutrophil assays and animal skin and sepsis models to analyze virulence of Panton-Valentine leukocidin (PVL)-positive and PVL-negative CA-MRSA strains as well as isogenic deletion knockouts of lukS/F-PV (endoding PVL) in strains USA300 and USA400. Findings from the study demonstrate that PVL is not the major virulence determinant of CA-MRSA.

This study linked production of cytolytic peptides (PSMs) with enhanced virulence of CA-MRSA. CA-MRSA strains showed increased production of PSMs and isogenic deletion of PSMα (in strains USA400 and USA300) significantly attenuated skin lesions in mice. This study also demonstrated that wild-type PSMs caused significant lysis of human neutrophils.

This study explored interfering with quorum sensing signally and subsequent virulence factor production as a means to ameliorate staphylococcal virulence. The study used an antibody specific to the autoinducing peptide (AIP) of the accessory gene regulator agr to demonstrate that interfering with the agr signaling mechanism disrupts normal agr function rendering S. aureus less virulent. This observation advances the search for potential novel antistaphylococcal therapeutics.
