    • Roles of Arabidopsis ATP/ADP isopentenyltransferases and tRNA isopentenyltransferases in cytokinin biosynthesis
    • This paper describes the isolation and characterization of mutants of the cytokinin biosynthesis genes, the ATP/ADP-isopentenyltransferases (AtIPTs). The ipt 1,3,5,7 multiple mutants showed a reduction in trans-zeatin and iP cytokinin content but not cis-zeatin, while ipt 2,9 mutants showed a decrease in cis-zeatin levels only, indicating that trans-zeatin and cis-zeatin have different biosynthetic pathways. Furthermore, ipt 1,3,5,7 mutants had longer roots and smaller shoots, suggesting a role for cytokinin during the development of these organs.

    • Direct control of shoot meristem activity by a cytokinin-activating enzyme
    • The authors discovered a novel component of the cytokinin biosynthesis pathway, the LONELY GUY (LOG) locus. The LOG gene encodes an enzyme able to convert an inactive cytokinin form into an active one. LOG is expressed in the central zone of the shoot apical meristem and the rice log mutant had a smaller shoot apical meristem and defects in flower meristem development, suggesting a key role for cytokinin in shoot apical meristem maintenance.

    • A subset of Arabidopsis AP2 transcription factors mediates cytokinin responses in concert with a two-component pathway
    • The authors have discovered a novel component of the cytokinin-signaling pathway: the CYTOKININ RESPONSE FACTORS (CRFs). CRFs are members of the AP2 family of transcription factors and are induced by cytokinin through the two-component signaling pathway. Moreover, cytokinins induce their rapid translocation to the nucleus. Analysis of crf null mutants revealed that these genes have overlapping functions in cytokinin response and operate in tandem with type-B ARRs, acting as cytokinin primary response genes.

    • Through the analysis of single, double, and triple mutants of the cytokinin receptors, the authors provide evidence that cytokinin has a role in various processes during plant growth and development. Furthermore, these data indicate that cytokinin receptors have partially redundant functions, and that cytokinins have opposite regulatory functions in shoot and root development.

    • Cytokinin signaling and its inhibitor AHP6 regulate cell fate during vascular development
    • The authors show that cytokinin phytohormones negatively regulate protoxylem specification. AHP6, an inhibitory pseudophosphotransfer protein, allow protoxylem formation antagonizing cytokinin signaling. On the contrary, cytokinin signaling negatively regulates the spatial domain of AHP6 expression. Thus, the circuitry between cytokinin signaling and AHP6 regulates proliferation and differentiation of cell lineages during vascular development by controlling the identity of cell lineages.

    • The RETINOBLASTOMA-RELATED gene regulates stem cell maintenance in Arabidopsis roots
    • This paper reports that local reduction of expression of the RETINOBLASTOMA-RELATED (RBR) gene in Arabidopsis roots increased the amount of stem cells without affecting the cell cycle. Conversely, overexpression of RBR-induced premature differentiation of the stem cell niche. An increase in KPR2 (cyclin D/kinase inhibitor) protein levels phenocopied the overexpression of RBR, while overexpression of cyclin D or E2F/DP had the same effect as decreased RBR levels, demonstrating that the shift from stem cell identity to differentiation is dependent on the canonical RBR pathway. Finally the authors provide evidence that RBR regulates stem cell identity acting downstream of the SCARECROW (SCR) gene.
