    • Size of supramolecular SNARE complex: membrane-directed self-assembly
    • In an earlier study, it had been demonstrated that membrane-associated t-SNAREs and membrane-associated v-SNARE interact in a circular array to form conducting channels in the presence of calcium, thus establishing continuity between opposing bilayers [7]. In the absence of membrane, no such SNARE-ring/channel complexes are formed. This study reports for the first time that the size of the SNARE-ring complex formed by the interaction of membrane-associated t-SNAREs and v-SNARE is directly proportional to the size of the vesicle the v-SNARE has been reconstituted in. Small proteoliposomes produce small SNARE-ring complexes, hence vesicle curvature regulates the size of the SNARE complex.

    • N-Ethylmaleimide-sensitive factor is a right-handed molecular motor
    • The SNARE complex is an SDS-resistant and an extremely stable complex. Exposure of t-/v-SNARE complex to the ATPase N-ethylmaleimide-sensitive factor (NSF) and ATP results in the disassembly of the complex, which can be imaged at near nanometer resolution using AFM. This study shows that t-/v-SNAREs form left-handed supercoils, which are uncoiled by NSF, thus demonstrating for the first time that NSF is a right-handed molecular motor.
