    • 4 cells/ml for HT-1080 and WI-38) were plated into 96-well microplates and were incubated for 24 h (for CCRF-CEM) or 4 h (for HT-1080 and WI-38). Various concentrations of nucleoside solutions (10 μl) in PBS containing 5% DMSO were then added into each well. The presence of 5% DMSO, which was used to dissolve the nucleosides, did not affect the cell growth (data not shown). After an incubation for 72 h, the MTT solution (25 μl) in PBS (5 mg/ml) was added to each well, and the plates were incubated for an additional 2 h. Finally, a 100 μl aliquot of a lysis solution (50% DMF/20% SDS) was added to each well, in order to dissolve the resulting MTT-formazan, and after an incubation for 16 h, the absorbance of the formazan product obtained by MTT reduction was measured at 570 nm, using a microplate reader.
