Using differential fluorescence induction, the authors identified 50 V. harveyi QS-regulated promoters and examined their responses to different combinations of autoinducers. They found that the various combinations of signals induce production of distinct levels of LuxR, which in turn determines the QS response at each target promoter. The promoters were divided into three classes corresponding to their affinities for LuxR. This regulatory circuit provides V. harveyi the potential to respond uniquely to different combinations of autoinducers. Importantly, it was demonstrated that the entire bacterial population responds in unison to the various autoinducer inputs.

The small RNA chaperone Hfq and multiple small RNAs control quorum sensing in Vibrio harveyi and Vibrio cholerae

Using a genetic screen to identify additional components of the V. harveyi and V. cholerae QS circuits, the authors identified the small RNA chaperone Hfq and showed that it mediates the destabilization of the mRNA encoding the master QS regulators LuxR (V. harveyi) and HapR (V. cholerae). Finding Hfq implicated sRNAs in the circuits, and using a bioinformatics approach, the authors identified four sRNAs in V. cholerae that function redundantly to control QS. Hfq, together with these sRNAs is proposed to create an ultrasensitive regulatory switch that controls the critical transition into the high cell density, QS mode. This provides the first demonstration of a role for sRNAs in QS in Gram-negative bacteria. It also uses an elegant combination of genetic, molecular and bioinformatic approaches.

The transcript levels of all the Qrrs was measured in different mutants, different growth stages, and in the presence of different autoinducers concentrations. The results presented show that the Qrrs levels can respond rapidly to changes in autoinducers inputs and therefore enable a rapid response to changes in cell density. The authors propose that in V. harveyi the multiple Qrrs are the key elements responsible for translating increasing autoinducers concentrations into a precise gradient of LuxR. LuxR, in turn, induces a gradient of expression of QS targets.

A genetic screen for V. cholerae QS regulators led to the identification of the VarS-VarA two-component sensory system that functions in parallel with the CAI-1 and AI-2 systems of V. cholerae. This third sensory system transmits information directly to LuxO, and unlike the other two systems, acts independently of LuxU. The VarS-VarA system is homologous to the GacS-GacA system that is present in many Gram-negative bacteria. In V. cholerae VarS-VarA controls the transcription of genes encoding three sRNAs - CsrB, CsrC and CsrD - which act redundantly to inhibit the activity of the global regulatory protein CsrA. Subsequently, CsrA regulates the phosphorylation state of LuxO either by increasing the level of phosphorylation of LuxO or the level of LuxO-P activity, resulting in inhibition of QS. The authors demonstrate that CsrBCD are equivalent to CsrB and CsrC of E. coli by complementation experiments carried out with the V. cholerae components expressed in E. coli.

Using transcriptome analysis of an hfq-null strain the authors demonstrate that Hfq affects approximately 5% of P. aeruginosa transcripts. Among these transcripts, some are also QS-regulated. Expression studies reveal that Hfq does not regulate the Las system, but does stimulate the translation of the rhlI gene indirectly by binding and stabilizing RsmY but not RsmZ. This finding suggests that these two sRNAs could be regulated by distinct factors. The authors present a model wherein the stabilization of RsmY alleviates the negative effect of RsmA, which accounts for the observed stimulation of rhlI expression by Hfq. It is also shown that Hfq is involved in the regulation of PQS and QscR, suggesting the existence of as yet unidentified Hfq-dependent sRNAs controlling QS in P. aeruginosa.

This study reveals that ExpR activates the expression of rsmA which encodes a global regulatory RNA-binding protein. The authors establish for the first time that this activation occurs through direct and specific interaction between ExpR and the rsmA promoter. Interestingly, when the AHL QS signal binds ExpR, it prevents the binding of ExpR to rsmA DNA, thus preventing the activation of rsmA transcription and subsequently allowing exoprotein and secondary metabolites production.

The authors demonstrate that the three sRNAs also positively regulate swarming motility and the synthesis of a low-molecular-weight QS signal, which is unrelated to AHLs and is produced and secreted in late exponential phase. Importantly this unknown signal induces the Gac-Rsm cascade and indicates that the GacS-GacA circuit of P. fluorescens might act as a QS system.
