Using mice possessing an inactivating MC1R mutation, the authors show that UV light fails to stimulate pigmentation in the absence of functional MC1R. This failure was rescued upon topical treatment with the cAMP agonist forskolin, emphasizing the role of MSH-MC1R signalling in the tanning response.
