Volume 75: Protein Evolution. Edited by Toone EA: Wiley & Sons; 2007:193-239; in press. This extensive review discusses evolution of enzyme superfamilies. In addition to mechanistically diverse superfamilies, it examines 'suprafamilies' in which substrate binding or other features of active site architecture have been conserved, but mechanistic attributes of catalysis have diverged. Also discussed are the role of promiscuity in protein evolution and how these evolutionary concepts might be applied to protein engineering.

    • Many enzymes in the Nudix superfamily have been structurally and mechanistically characterized, and this review pulls this information together to describe the conserved and divergent aspects of catalysis. Detailed comparisons of the mechanisms of four Nudix proteins are used to derive mechanistic concepts that are applied to less well-characterized superfamily members.
