Hum Mutat 2005; 25:566-574. Germline RB1 mutations were identified in 77 out of 85 bilateral retinoblastoma patients (91%), seven out of 10 familial unilateral (70%), and six out of 85 unilateral patients with no family history of retinoblastoma (7%). Mutations included 36 novel alterations spanning the entire RB1 gene.

There were 158 eyes with retinoblastoma treated with chemoreduction. The eyes were grouped based on a simple classification: group 1, tumor only; group 2, tumor plus subretinal fluid; group 3, tumor plus focal seeds; group 4, tumor plus diffuse seeds; and group 5, neovascular glaucoma or invasive retinoblastoma. The classification system predicted success with chemoreduction as there was 100% success for group 1, 91% for group 2, 59% for group 3, and 12% for group 4 (group 5 was always managed by primary enucleation).
