    • There are two possible explanations for the effect exerted by conformational dynamics on the fluorescent signal. The first builds on the changes of the stochastic frequency and amplitude profile of intramolecular movements caused by covalent substitution, whereas the other advocates a change in the static equilibrium structure accompanying the supramolecular modification. At present, we presume that simple sensor molecules are better described by the first approach, whereas for the characterization of supramolecular complexes, the second approach might be more relevant. Experimental and modeling studies aimed at refining this picture are in progress.
