    • The (DMPC/cholesterol) mixed monolayer with a molar fraction of 70/30 on water had been characterized to be in a phase-separated state by a fluorescence microscope. There is a difference in content of cholesterol (also DMPC) between domain and matrix regions in the monolayer. The molayer observation on aqueous VCH monolayer. The monolayer observation on aqueous VCH may offer the key to specify which component of the monolayer is responsible for binding with VCH. Then, we adopted the molar fraction of 70/30 as the condition of monolayer preparation.

    • The π-A isotherm of the mixed monolayer on aqueous VCH solution was measured by changing the standing time up to 90 min. The isotherm slightly shifted to a larger surface area even at 90 min. This indicates that more than 90 min is required for the adsorption saturation of VCH. However, it is difficult to measure the isotherm for long time because of water evaporation in subphase [T. Kuri, et al., Langmuir 13 (1997), 6497]. Then, we adopted 30 min as the adsorption time of VCH in this study.

    • The color contrast of the magnified AFM image was emphasized in comparison with the height difference. The average surface roughness of the magnified AFM image was 0.11 nm.

    • Molecular weight of VCH was estimated to be 65 kDa by SDS-polyacrylamide gel electrophoresis. The dot in a matrix region may correspond to an aggregate, composed of 4 or 5 subunits, of VCH.
