Mixed active site mutants of RNase A can partially recover their enzymatic activity through oligomerization via 3D domain swapping. The authors used this recovery of activity to show that fibrils of RNase A polyglutamine mutants form via 3D domain swapping. This study shows that proteins need not lose their native structure to form amyloid-like fibrils and that 3D domain swapping can account (at least in part) for the protein self-specificity of amyloid-like fibrils.
