    • Using a range of biophysical techniques, the authors show that the prion domain of HET-s forms four β-strands in fibrils; these strands are arranged into two strand-turn-strand structures that are probably parallel stacked. This first medium to high resolution model proposed for HET-s fibrils sheds light on the structure of an 'evolved' amyloid-like fibril.

The study demonstrates the power of solid-state NMR as a tool for examining amyloid fibril structure and confirms the proposal that Ure2p forms parallel β-sheets with a polar zipper of Gln18 sidechains.

The peptides stack parallel and in register into a cross-β spine arrangement in which two sheets pack together in a very tight, self-complementary steric zipper. This is the first publication to present atomic-level structural details of an amyloid-like cross-β spine.

    • On the basis of a previously published X-ray diffraction pattern for polyglutamine peptide assemblies, the authors propose a model in which the peptides stack in flat antiparallel β-sheets, and the glutamine sidechains form hydrogen-bonded stacks within a sheet and interdigitate tightly between sheets. This paper is an excellent example of careful and thorough interpretation of semi-crystalline diffraction data (which is also applicable to fiber diffraction data).
