Cross-talk between the cytosolic mevalonate and the plastidial methylerythritol phosphate pathways in tobacco bright yellow-2 cells

A. Hemmerlin, J.F. Hoeffler, O. Meyer, D. Tritsch, I.A. Kagan, C. Grosdemange-Billiard, M. Rohmer, and T.J. Bach Cross-talk between the cytosolic mevalonate and the plastidial methylerythritol phosphate pathways in tobacco bright yellow-2 cells J Biol Chem 278 2003 26666 26676 The authors show, using stable isotope-labeled precursors and specific inhibitors, that there is crosstalk between the MVA and MEP pathways in tobacco BY-2 cell suspensions. Apparently, in these cells, there is transport of MVA intermediates from the cytosol to the plastid, as well as transport of MEP intermediates from the plastid to the cytosol. The exact intermediates are not known, but this paper indicates that, under certain conditions, transport might be bidirectional.

Formation of monoterpenes in Antirrhinum majus and Clarkia breweri flowers involves heterodimeric geranyl diphosphate synthases

D. Tholl, C.M. Kish, I. Orlova, D. Sherman, J. Gershenzon, E. Pichersky, and N. Dudareva Formation of monoterpenes in Antirrhinum majus and Clarkia breweri flowers involves heterodimeric geranyl diphosphate synthases Plant Cell 16 2004 977 992 The authors show that the monoterpene-emitting flowers of A. majus and C. breweri contain a small and a large GPS subunit that are only active as a heterodimer. The expression of the small subunit correlates closely with monoterpene emission, indicating that it plays a regulatory role in monoterpene formation.

N. Dudareva, S. Andersson, I. Orlova, N. Gatto, M. Reichelt, D. Rhodes, W. Boland, and J. Gershenzon The nonmevalonate pathway supports both monoterpene and sesquiterpene formation in snapdragon flowers Proc Natl Acad Sci USA 102 2005 933 938 Using stable isotope-labeled precursors and specific inhibitors, the authors show that the plastidial MEP pathway, and not the cytosolic MVA pathway, provides precursors for both the plastidial monoterpene synthases and for the cytosolic sesquiterpene synthase. Moreover, the diurnal rhythm of the MEP pathway controls the diurnal emission of terpenoids, providing evidence for precursor control of terpenoid emission.

Characterization of the Arabidopsis clb6 mutant illustrates the importance of posttranscriptional regulation of the methyl-d-erythritol 4-phosphate pathway
-
A. Guevara-Garcia, C. San Roman, A. Arroyo, M.E. Cortes, M. de la Luz Gutierrez-Nava, and P. Leon Characterization of the Arabidopsis clb6 mutant illustrates the importance of posttranscriptional regulation of the methyl-d-erythritol 4-phosphate pathway Plant Cell 17 2005 628 643 The authors describe an albino Arabidopsis mutant that carries a defect in the gene that encodes the last enzyme in the MEP pathway. Interestingly, this mutant reveals that the first enzyme in the pathway, DXS, is feedback regulated and that some of the MEP pathway proteins are post-transcriptionally regulated. This study indicates that there is still a lot is to learn about the regulation of the MEP pathway.

Characterization of GaWRKY1, a cotton transcription factor that regulates the sesquiterpene synthase gene (+)-delta-cadinene synthase-A
-
Y.H. Xu, J.W. Wang, S. Wang, J.Y. Wang, and X.Y. Chen Characterization of GaWRKY1, a cotton transcription factor that regulates the sesquiterpene synthase gene (+)-delta-cadinene synthase-A Plant Physiol 135 2004 507 515 The authors isolated a WRKY transcription factor that specifically binds to the cadinene synthase promoter. This WRKY interacts with the promoter in a yeast one-hybrid system and is capable of activating the promoter in tobacco leaves and in transgenic Arabidopsis. This study provides new insights into how terpene synthases in plants can be regulated.
