These are the first studies to investigate the molecular cross-talk that occurs between cell types present in the hematopoietic microenvironment. Genomewide microarray expression profiling was used effectively to dissect the microenvironmental cross-talk. The authors provide further evidence for the importance of Notch signaling in hematopoiesis.

    • Visnjic D, Kalajzic Z, Rowe DW, et al.: Hematopoiesis is severely altered in mice with an induced osteoblast deficiency. Blood 2004, 103:3258-3264. A transgenic model is used to ablate osteoblasts specifically, causing a dysregulation of hematopoiesis in the marrow but a concordant rise in extramedullary hematopoiesis. This is an inverse but complementary approach to the models used by others [29••,30••] to demonstrate the importance of osteoblasts in the stem cell niche.

    • Wnt proteins are lipid-modified and can act as stem cell growth factors
    • Willert K, Brown JD, Danenberg E, et al.: Wnt proteins are lipid-modified and can act as stem cell growth factors. Nature 2003, 423:448-452. This is the first report of a purified Wnt protein that has growth factor activity on HSC. The study demonstrates of expansion of repopulating stem cells with only Wnt 3a protein and minimal amounts of stem cell factor.
