Characteristic white colonies on MacConkey agar were used to inoculate triple sugar iron (product no. 211749, Becton Dickinson and Company, Sparks, MD) and lysine iron agar slants (product no. 284920, Difco Laboratories), which were incubated at 37°C for 24 h.

The cells were suspended in 200 mL of sample buffer via vortexing. A 30-mL aliquot of this sample was transferred to a single well of an 8-well sample carrier. The sample carrier, buffer, and all further reagents were part of a kit designed for the RiboPrinter System. The restriction enzyme PvuII was used to differentiate ribotypes.
