The bHLH transcription factor Twist is identified as a gene upregulated early during metastatic progression, and is functionally validated as a critical regulator of metastasis, using short interfering RNAs. Twist is able to promote EMT in mammary epithelial cells, in part via downregulation of E-cadherin. The high levels of Twist expression seen in 70% of invasive lobular breast carcinomas, which display many features of EMT, are inversely correlated with E-cadherin expression. Identification of Twist provides a powerful tool for studying the role of EMT during tumor metastasis, in particular during the movement of cells from a primary tumor into the circulation.

Ha-Ras-transformed mammary epithelial cells, which undergo EMT in response to TGFβ, are prevented from progressing through EMT when NF-κB activity is blocked. NF-κB pathway activation, however, promotes EMT even without TGFβ. Moreover, NF-κB inhibition reverses EMT in mesenchymal cells. Inhibiting the NF-κB pathway in mice injected with the Ha-Ras-transformed mammary epithelial cells abrogates the metastatic potential of these cells. Collectively, these data provide evidence that NF-κB plays an essential role in both the induction and maintenance of EMT.

A Snail variant that is non-phosphorylatable at both sites is sufficient to cause EMT, and a correlation is observed between inactive, phosphorylated GSK-3 and Snail expression in human breast carcinomas.
