Jensen Mmm2p, a mitochondrial outer membrane protein required for yeast mitochondrial shape and maintenance of mtDNA nucleoids J Cell Biol 164 2004 677 688 Using a synthetic lethal scheme, the authors identify Mmm2p, a protein that appears to interact with Mmm1p. Like Mmm1p, the outer membrane Mmm2 protein is found in dot-like structures next to mtDNA nucleoids, and is required for normal mitochondrial shape. In vivo studies suggest that the Mmm1p- and Mmm2p-containing particles dynamically interact.

Immunofluorescence and immunoprecipitation analysis indicates that Mmm1p, Mdm10p and Mdm12p interact, raising the possibility that they are all part of the same structure.

SAM is part of the protein import machinery used to insert β-barrel proteins in the outer membrane. Also blurring the distinction between import and shape, the authors find that defects in two known import components, Tom40 and Sam37p, lead to altered mitochondrial morphology.
