By the use of several delivery systems, recombinant L. lactis strains could be designed to target heterologous proteins to the cell cytoplasm, the cell wall and to the supernatant of the growth medium of the bacterium. The authors describe an application of the use of recombinant L. lactis as an antigen delivery vehicle for the development of live mucosal vaccines.

The authors compare this element with four other LAB elements that are related to it. ICESt1 contains a recombination site. This element might be excised from the genome by a site-specific recombination event between two direct repeat sequences in its two attachment sites, which leads to the excision of the circular molecule. The element possesses ORFs encoding conjugative functions and it also contains a putative origin of transfer (oriT).

The recombinant L. lactis strains with vectors that contained the vp2 and those with the vp3 genes that were intended to target these antigens to the cell wall and the supernatant of the growth medium, efficiently targeted VP3 to these designated locations. This was not so for L. lactis with the secretion vector that contained vp2, and VP2 failed to be secreted. Importantly, L. lactis with the secretion vector with nuc-vp2 and that with the vector with nuc-vp2-cwa brought about export of VP2 to the supernatant and to the cell wall, respectively. Furthermore, the administration of cells of the strain with the secretion vector with nuc-vp2-cwa to chickens induced an IgG response against Nuc, but the authors did not detect any serum IgG response against VP2.

BALB/c mice administered orally with cells of IL1403-pHIV showed that the immunised mice contained 300-fold more HIV-specific serum IgG and 100-fold more fecal IgA than mice with IL1403-pEmpty. Assays of the cells of the spleen and the peritoneal lymph nodes of immunised mice to detect HIV-specific cell-mediated immunity showed that the number of HIV-specific IFN-γ-secreting cells in IL1403-pHIV-immunised mice was significantly greater than in controls. Importantly, when vaccinated mice were intraperitoneally challenged with vaccinia virus vPE16 (expresses the HIV env gene) this reduced viral load by 350-fold.
