Howe The tomato homolog of CORONATINE-INSENSITIVE1 is required for the maternal control of seed maturation, jasmonate-signaled defense responses, and glandular trichome development Plant Cell 16 2004 126 143 The work described in this paper demonstrates that the tomato jai1-1 mutant, which is impaired in recognition of the mobile wound signal, harbors a null mutation in the tomato homolog of COI1. The phenotype of jai1 plants confirms the importance of the JA-COI1 pathway in herbivore defense, and further reveals novel roles for jasmonates in maternal control of seed maturation and glandular trichome development.

Kobayashi Role of β-oxidation in jasmonate biosynthesis and systemic wound signaling in tomato Plant Cell 17 2005 971 986 The authors used a map-based cloning approach to identify an isoform of acyl-CoA oxidase (ACX1A) that plays an essential role in the β-oxidation stage of JA biosynthesis. The characterization of acx1 mutant plants showed that JA, rather than a C18 precursor of JA, is required for induced defense against a chewing insect, and for the production of a mobile wound signal that activates systemic defenses. These findings establish an essential role for peroxisomes in the production of a JA-based systemic signal.

Ryan The cellular localization of prosystemin: a functional role for phloem parenchyma in systemic wound signaling Planta 218 2004 360 369 The authors show that prosystemin, the precursor of systemin, is expressed in the phloem parenchyma cells of tomato and potato. This finding indicates that vascular parenchyma cells are directly involved in systemic wound signaling. Taken together with the results of other studies showing that JA-biosynthetic enzymes are located in the companion cell-sieve tube element complex [40], this work supports a model in which (pro)systemin acts to induce the production of a phloem-mobile jasmonate signal.

    • Effects of feeding Spodoptera littoralis on lima bean leaves. II. Continuous mechanical wounding resembling insect feeding is sufficient to elicit herbivory-related volatile emission
    • A. Mithofer, G. Wanner, and W. Boland Effects of feeding Spodoptera littoralis on lima bean leaves. II. Continuous mechanical wounding resembling insect feeding is sufficient to elicit herbivory-related volatile emission Plant Physiol 137 2005 1160 1168 The authors of this important paper used a 'mechanical caterpillar' to simulate herbivore feeding. Their results show that mechanical wounding, if administered in a way that closely resembles the insect's feeding process, is sufficient to induce a systemic defense response that is qualitatively similar to that induced by insect feeding. These results indicate that the contribution of mechanical wounding to the activation of herbivore-induced host defense responses is greater than previously realized.

    • The oxylipin signal jasmonic acid is activated by an enzyme that conjugates it to isoleucine in Arabidopsis
    • P.E. Staswick, and I. Tiryaki The oxylipin signal jasmonic acid is activated by an enzyme that conjugates it to isoleucine in Arabidopsis Plant Cell 16 2004 2117 2127 The authors demonstrate that the primary biochemical function of the JAR1 enzyme in Arabidopsis is the conjugation of JA to isoleucine. On the basis of the reduced responsiveness of jar1 mutant plants to JA, their results demonstrate that the conjugation reaction confers new signaling properties to JA.

    • Structural and biochemical studies identify tobacco SABP2 as a methyl salicylate esterase and implicate it in plant innate immunity
    • F. Forouhar, Y. Yang, D. Kumar, Y. Chen, E. Fridman, S.W. Park, Y. Chiang, T.B. Acton, G.T. Montelione, and E. Pichersky Structural and biochemical studies identify tobacco SABP2 as a methyl salicylate esterase and implicate it in plant innate immunity Proc Natl Acad Sci USA 102 2005 1773 1778 The authors show that SABP2 functions as a methyl-SA esterase. This result, together with previous work showing that silencing of SABP2 abrogates systemic acquired resistance (SAR) to pathogens, suggests that the role of SABP2 in SAR is to convert inactive methyl-SA to an active pool of SA that subsequently mediates defense gene expression.
