XLPOU-60 protein, translated from a transcript encoding a c-myc-tagged version of XLPOU-60, is shown to undergo a change in its cellular distribution from cytoplasmic to nuclear, coincident with onset of transcription of the zygotic genome.

Development 1996, 122:881-894. The authors have used transgenic mapping to identify the two enhancers required for germ cell and epiblast expression of Oct-3/4. In vitro expression directed by the epiblast enhancer is downregulated during retinoic acid-mediated differentiation of P19 cells, suggesting that retinoic acid present in Hensen's node may function in the downregulation of Oct-3/4 in vivo.

    • Bermingham JR Jr, Scherer SS, O'Connell S, Arroyo E, Kalla KA, Powell FL, Rosenfeld MG: Tst-1/Oct-6/SCIP regulates a unique step in peripheral myelination and is required for normal respiration. Genes Dev 1996, 10:1751-1762. The authors have used targeted gene replacement to generate mice that are null for the Tst-1/Oct-6/SCIP locus. These mice die shortly after birth because of a breathing defect. Disruption of peripheral but not of central myelination is observed in Tst-1/Oct-6/SCIP null mice, yet the expression of myelin structural genes appears unaffected. The data suggest that Tst-1/Oct-6/SCIP regulates a unique step in peripheral myelination.

    • Schonemann MD, Ryan AK, McEvilly RJ, O'Connell SM, Arias CA, Kalla KA, Li P, Sawchenko PE, Rosenfeld MG: Development and survival of the endocrine hypothalamus and posterior pituitary gland requires the neuronal POU domain factor Brn-2. Genes Dev 1995, 9:3122-3135. The authors have created a Brn-2 null mouse by targeted disruption of the locus. Brn-2 null mice are severely runted and die within 10 days of birth. The mutants exhibit extensive hypocellularity of their supraoptic and paraventricular nuclei and failure of axonal projections that results in loss of the posterior pituitary gland. Early events in neurogenesis do not appear to be affected. The stratified expression pattern of class III POU domain proteins in the developing hypothalamus is also described.

A localized requirement for nubbin in the hinge region seems to be involved in growth and patterning of the wing.

    • On the functional overlap between two Drosophila POU homeo domain genes and the cell fate specification of a CNS neural precursor
    • Yeo SL, Lloyd A, Kozak K, Dinh A, Dick T, Yang X, Sakonju S, Chia W: On the functional overlap between two Drosophila POU homeo domain genes and the cell fate specification of a CNS neural precursor. Genes Dev 1995, 9:1223-1236. The closely linked and structurally related POU domain proteins, pdm-1 and pdm-2, are essential for specifying the identity of the ganglion mother cell GMC4-2a and production of the RP2 neuroblast lineage. pdm-2 is the dominant factor in this process.

    • The miti-mere and pdm1 genes collaborate during specification of the RP2/sib lineage in Drosophila neurogenesis
    • Bhat KM, Poole SJ, Schedl P: The miti-mere and pdm1 genes collaborate during specification of the RP2/sib lineage in Drosophila neurogenesis. Mol Cell Biol 1995, 15:4052-4063. Characterization of the role of pdm-1 and miti-mere (pdm-2) in ganglion mother cell (GMC-1) identity. Both genes are required for the normal development and correct specification of this lineage. Overexpression of either pdm-1 or miti-mere results in duplication of RP2 motoneurons.
