Muday The transparent testa4 mutation prevents flavonoid synthesis and alters auxin transport and the response of Arabidopsis roots to gravity and light Plant Cell 16 2004 1191 1205 This study examines several effects of endogenous flavonoids (or the lack thereof) on auxin distribution in the root during gravitropic and light responses. It argues that environmental stimuli influence flavonoid accumulation, which in turn regulates auxin transport. Interestingly, the gravitropic response in tt4 roots was not abolished, only delayed, compared to wild-type; moreover, the delay was biochemically complemented with naringenin.

Murphy Variation in expression and protein localization of the PIN family of auxin efflux facilitator proteins in flavonoid mutants with altered auxin transport in Arabidopsis thaliana Plant Cell 16 2004 1898 1911 By using flavonoid-deficient and flavonoid-overexpressing mutants, this study shows that flavonoids inhibit auxin transport primarily at the shoot apex and the root tip. Flavonoids do not directly affect the expression of various PIN genes, but they might modulate PIN cycling in the root cells. The authors interpret the data as consistent with a role for flavonoids in regulating the phosphorylation of vesicular trafficking proteins.

This study demonstrates that transgenic pollen that expresses an antisense copy of the SHY cDNA exhibits an abnormality in tube growth that is more pronounced in vivo than in vitro. The generation of large callose deposits and growth arrest of the pollen tube in the pistil lead to functional male sterility.

    • H. Kobayashi, Y. Naciri-Graven, W.J. Broughton, and X. Perret Flavanoids induce temporal shifts in gene-expression of nod-box controlled loci in Rhizobium sp. NGR234 Mol Microbiol 51 2004 335 347 This study measured the NodD-dependent response to flavonoids of the 19 nod-boxes present within the symbiotic plasmid of the broad host-range Rhizobium sp., NGR234, to flavonoids. Nine flavonoids were tested as inducers of nod-boxes and the transcriptional response was measured in both wild-type and regulatory mutant backgrounds. Of the 19 nod-boxes tested, 18 were found to be flavonoid-inducible and showed temporal and strength differences in their transcriptional response. The authors present a three-tiered model of the flavonoid-dependent regulatory cascade that sequentially activates the expression of 75 loci involved in Nod-factor synthesis, nodule initiation, a type III secretion system, and synthesis of late-stage infection factors.

    • Flavonoid binding to a multi-drug-resistance transporter protein: An STD-NMR study
    • L. Nissler, R. Gebhardt, and S. Berger Flavonoid binding to a multi-drug-resistance transporter protein: an STD-NMR study Anal Bioanal Chem 379 2004 1045 1049 This study shows that substitutions at carbon 5 (C5) that modify the polar group result in reduced binding and activity. This finding agrees with an earlier study [58] that showed that MDR function is enhanced by an unsubstituted OH at position C5. The two studies disagree on the basis of this action: the NMR data suggest it is owing to increased lipophilicity to the flavonoid nucleus from the intramolecular bridging between the C5 OH and the adjacent ketone at C4.
