14. 2 atmosphere. An aliquot (200 μL) of the medium containing test sample was added to the each well, and the cells were cultured for 20 h. The plate was centrifuged (2000 rpm, 4°C, 10 min), the supernatant was removed, and 120 μL of distilled water was added to each well. The hepatocytes were broken with sonication, and the suspension was centrifuged. To determine the concentration of triglyceride in the supernatant using a commercial kit (Triglyceride G-test wako), each test compound was dissolved in DMSO, and the solution was added to the medium (final DMSO concentration was 0.5%).
